MC-GGFG-Exatecan

It consists of Exatecan, a highly potent topoisomerase I inhibitor (cytotoxic payload), attached through a MC-GGFG peptide linker (maleimidocaproyl–glycine-glycine-phenylalanine-glycine). The MC group enables stable conjugation to cysteine residues on antibodies, while the GGFG peptide linker is designed to be selectively cleaved by lysosomal enzymes inside target cells. Once internalized by cancer cells, the linker is enzymatically degraded, releasing active Exatecan to induce DNA damage and cell death. This design improves targeted delivery, enhances therapeutic efficacy, and reduces systemic toxicity compared to conventional chemotherapy. MC-GGFG-Exatecan is a cleavable linker–payload complex often used in next-generation antibody-drug conjugates (ADCs).
